SH3 and multiple ankyrin repeat domains protein 2 (SHANK2) is a member of the Shank gene family known as A0081. Other members of the Shank gene family are post-synaptic proteins containing multiple domains including ankyrin repeats, an SH3 domain, a PSD95/Dlg/ZO-1 domain, a sterile alpha motif domain, and a proline-rich region. In particular, Shank2 contains a PDZ domain, which binds to the Src homology 3 (SH3) domain of cortactin. Shank2 is suggested to function as an adapter protein in the post-synaptic density of excitatory synapses. It probably plays a role in the organization of the dendritic spine and synaptic junction. Shank2 is involved in a complex with PSD95/DLG4 and DLGAP1/GKAp. It also interacts with other synaptic proteins such as GRID2 and DNM1.
